Effect of Medium Composition on Multiple Shooting and Subsequent Growth of Mericlone from Rhizome of Shoot Tip Cultures of Temperate Cymbidium Species




Abstract
This experiments were conducted in order to get the optimum level of plant growth regulators in every culture stage for ultimately obtaining well established mericlones from shoot tip derived rhizomes of temperate Cymbidium species. Multiple shooting from rhizomes of C. forrestii ¢¥Sojub¢¥ and ¢¥Nokwoon¢¥, and C. karan ¢¥Soshim¢¥ was enhanced in the medium supplemented with 1.0 §·¡¤L^(-1) NAA and 3.0 §·¡¤L^(-1) BA, but C. forrestii ¢¥Changhaso¢¥ produced numereous shoots in the medium added with 1.0 §·¡¤L^(-1) and 1.0 §·¡¤L^(-1) BA. On the other hand, C. nishiuchianum ¢¥Hoduchjiwha¢¥ were much more shouted, in the medium containing 2.0 §·¡¤L^(-1) NAA and 1.0 §·¡¤L^(-1) BA. When young shoots, within 5 to 6 §¯ in length, which were obtained in multiple shooting medium, were subcultured, growth of C. forrestii ¢¥Sojub¢¥ were markedly promoted in the medium added with 1.0 §·¡¤L^(-1) NAA and BA respectively, but C. forrestii ¢¥Songmae¢¥ and ¢¥Yangshiso¢¥ grew more rapidly in the medium enriched with 2.0 §·¡¤L^(-1) NAA and 1.0 §·¡¤L^(-1) BA. On the other hand, C. forrestii ¢¥Changhaso¢¥, C. karan ¢¥Soshim¢¥ and C. niveo-marginatum kept good growing in the medium containing 0.5 §·¡¤L^(-1) NAA and BA respectively. Subsequent shoot growth, in pretransplanting medium, of C. forrestii ¢¥Sojub¢¥ and ¢¥Songmae¢¥ were stimulated in 3 g¡¤L^(-1) Hyponex and 2 g¡¤L^(-1) peptone (H©ýP©ü) medium supplemented with 2.0 §·¡¤L^(-1) NAA and 1.0 §·¡¤L^(-1) BA and 500 §·¡¤L^(-1) activated charcoal, but the growth of C. forrestii ¢¥Changhaso¢¥ and C. karan ¢¥Soshim¢¥ in H©ýP©ü medium enriched with 1.0 §·¡¤L^(-1) NAA and BA, C. forrestii ¢¥Yangsiso¢¥ and C. niveo-marginatum in H©ýP©ü medium added with 2.0 §·¡¤L^(-1) NAA, 1.0 §·/L BA and 500 §·¡¤L^(-1) activated charcoal, and C. forrestii ¢¥Seosinmae¢¥ in 3 g¡¤L^(-1) Hyponex and 4 g/L peptone (H©ýP©þ) medium containing 2.0 §·¡¤L^(-1) NAA, 1.0 §·¡¤L^(-1) BA and 500 §·¡¤L^(-1) activated charcoal were more promoted compared with another compositions of the media.
